My child is anxious, and hasn't talked to peers in playgroup, but does participate. DC apparently mostly kept to themselves during playtime etc. We are in play therapy, which isn't helping much.
DC is now starting preschool in a new school. I some to the director a while ago about the diagnosises, so they are aware
When should I speak to the moros? Before the first day of school so I can ask them to keep an eye out for DC who is anxious about starting school? Or give DC a chance to make their own impressions on the teachers, and call them in a few days?
What do you all think?

Give it a few days.
You can say s/he is a little nervous and it might take some time for her/him to get comfortable.
The director might have also let the Morah know.

I feel like if you have a diagnosis , and you know how your child will present then why not be proactive to give your child the best chance on transitioning to a new Morah/school/friends…I would surely speak to the teacher if I can . Another idea is to ask the head of the school who knows ur situation if they recommend you speak to the teacher before especially since your child is an anxious child

Does she have selective mutism? If so you really should tell the teacher
