We are looking into a Lakewood move but DH is an accountant in NYC with late hours. He does have some flexibility to work from home. Is it doable?

It's definitely hard if he's also working long hours.
NYC - you don't mean Brooklyn, you mean Manhatttan?
It's a long trip with traffic, two hours maybe more.

Yes NYC
Are there buses that travel very late? Is it costly?

NJ transit goes, but how late are you looking? I know they definitely go from NYC to Lakewood area around 8pm, maybe even later..
There is a Kollel bus in the mornings.

From Lakewood to Manhattan on the bus often takes an hour and fifteen minutes.
Getting to and from the bus, on the other hand... can add time.
